Histochemical Fiber Types of Mammalian Extraocular Muscle, an Investigation of Medial Rectus Muscle


Abstract
This experiment was designed for the evaluation of muscle fiber types and characteristics of mammalian extraocular medial rectos muscles.
Flash-frozen muscle sections of extraocular medial rectos from cats, rabbits, rats and mice were stained for H-E, trichrome, PAS, regular and reversed myosin ATPase and succinate dehydrogenate(SDH), and examined microscopically.
The results obtained were as follows.
1. High enzyme activities of myosin ATPase and SDH with increased PAS reaction were noted in medial rectos muscles compared to those of tibias anterior muscles of mammals.
2. Two distinct global and orbital layers were evident and three muscle fiber types were classified in H-E, trichrome and PAS stained sections of medial rectos muscles.
3. Two fiber types (type I and II) were only identified and the predominant muscle fiber type If by regular myosin ATPase stain in global and orbital layers of mammalian medial rectus muscles.
4. Three muscle fiber types(type I, IIa and IIb) were differentiated with reversed myosin ATPase and SDH stains. Species and zonal(global and orbital) difference was markedly evident in the proportions of muscle fiber types, compared to little difference of regional (proximal, middle and distal) one.
5. Activities of myosin ATPase and SDH, and PAS reaction were more marked in muscle fibers of orbital layer, than those in global layer of mammalian medial rectos muscles.
6. Evaluation for the combination patterns of various enzyme histochemical activities in medial rectos muscles showed that immature type IIc and unusual muscle fibers were unexplainably abundant in number.
